Pinelloside, an antimicrobial cerebroside from Pinellia ternata   总被引:3,自引:0,他引:3

An antimicrobial cerebroside, pinelloside, was isolated from the air-dried tubers of Pinellia ternata (Thunb.) Breit. Its structure was determined as 1-O-beta-D-glucopyranosyl-(2S,3R,4E,11E)-2-(2'R-hydroxyhexadecenoylamino)-4,11-octadecadiene-1,3-diol by chemical transformation and extensive spectroscopic analyses (IR, MS, 1H and 13C NMR, DEPT as well as 2D NMR techniques HMBC, HMQC, 1H-1H COSY and NOESY). The antimicrobial assay showed that this compound was inhibitory to the growth of Bacillus subtilis, Staphylococcus aureus, Aspergillus niger and Candida albicans, with minimum inhibitory concentrations (MICs) of 20, 50, 30 and 10 microg/ml, respectively. The MICs of penicillin G against bacteria B. subtilis, S. aureus, E. coli, P. fluorescens and H. pylori were 0.80, 0.34, 0.56, 1.34 and 0.92, and those of ketoconazole against fungi A. niger, C. albicans and T. rubrum 0.90, 0.65 and 1.0 microg/ml, respectively.  相似文献

Pinellia ternata , a traditional Chinese herb that has been used in China for over 1000 years, is susceptible to a soft rot disease, which may cause major loss of yield. The use of bacteria as potential antagonists against Pectobacterium carotovorum SXR1, the causal agent of the disease on P. ternata , was evaluated. Altogether, 1107 candidate bacteria were isolated from the rhizosphere and surface-sterilized plants of P. ternata . In Petri dish tests, 55 isolates inhibited the growth of strain SXR1, and 21 of these reduced the disease development on P. ternata slices by over 50%. Four selected antagonists significantly reduced the disease incidence on tissue culture seedlings, and also prevented the disease on the transplants. Agonist P-Y2-2 yielded a good prevention level of 81.9%. The four antagonists rapidly colonized the tissue culture seedlings and transplants, whereas greater populations of the antagonists (107–109 CFU g−1 fresh tissues) were observed in the seedlings and in the preinoculated transplants than in those inoculated during transplanting. The use of pathogen-free tissue culture seedlings pre-inoculated with antagonist may provide a strategy for production of P. ternata plantlets resistant to soft rot disease. This is the first report on the efficacy of biocontrol agents against pathogens on P. ternata .  相似文献

The objective of this study was to determine the effect of two endophytic bacterial elicitors (Pseudomonas sp. and Enterobacter sp.) on the production of alkaloids in protocorm-like bodies (PLBs) of Pinellia ternata Breit. Both bacterial strains increased the growth rate of P. ternata PLBs. Pseudomonas sp. promoted the differentiation of the PLBs, whereas Enterobacter sp. inhibited PLB differentiation. The bacterial strains increased guanosine production in PLBs by 9–166%, inosine production by 2–33%, and trigonelline production by 114–1140% compared to the control. For Pseudomonas sp., guanosine and trigonelline production was greater when bacterial extracts were added to the PLB suspension cultures rather than living cells (co-culture treatment). Inosine production was similar in both the bacterial extract and co-culture treatments. For the Enterobacter sp., guanosine, inosine, and trigonelline production tended to be greatest when living cells were added to the PLB suspension cultures rather than bacterial extracts. These results suggest that Pseudomonas sp. and Enterobacter sp. could increase alkaloid yield from P. ternata under field or tissue culture conditions. We also observed that Pseudomonas sp. and Enterobacter sp. produced some of the same alkaloids as their host plants. Additional study needs to be done to determine if these endophytic bacteria could be used to produce alkaloids in the fermentation industry.  相似文献

以药用植物三叶半夏叶片为材料,通过比较直接和间接器官发生两种途径,建立了半夏一步成苗的快速繁殖技术体系。结果表明,经过愈伤组织阶段的一步成苗培养基为MS+0.5mg/L2,4-D+1.0mg/LKT,90d左右方可得到再生植株,植株分化率为74%,每个外植体上分化的块茎数为5.61±1.04。附加NAA与BA两种激素对一步成苗培养基进行优化,筛选出一步成苗最佳培养基MS+0.5mg/LNAA+0.5mg/LBA,60d后就可直接发育成完整植株,植株分化率为76%,每个外植体上分化的块茎数高达9.97±0·81,对这种培养基上的再生小植株进行移栽,1个月后,移栽成活率达100%。  相似文献

【目的】对贵州省半夏块茎腐烂病病原菌进行分离和鉴定,为开发有效的防治技术体系提供依据。【方法】采用组织分离法进行病原菌分离,根据柯氏法则进行致病性测定,结合形态学、生理生化特性和分子生物学方法进行鉴定。【结果】从贵州3个半夏产地患病样品中分离出12株病原细菌和5株病原真菌。经形态学、生理生化特性和分子生物学方法鉴定,12株病原细菌为胡萝卜软腐果胶杆菌胡萝卜软腐亚种;ZJ、Z3病原真菌为尖孢镰刀菌,D3、D5、H1为茄病镰刀菌。【结论】贵州省半夏块茎腐烂病的病原菌有病原细菌和病原真菌。  相似文献

目的 探讨半夏总生物碱(total alkaloids from Pinellia Ternate,TAPT)对帕金森病模型大鼠的防治作用及其抗氧化机制.方法 采用脑内定位注射6-羟基多巴胺(6-OHDA)建立帕金森病大鼠模型,在模型建立成功的同时给予半夏总生物碱预防性治疗.采用Morris水迷宫进行帕金森病大鼠的行为学检测,用化学比色法检测大脑皮质及血清超氧化物歧化酶(SOD)活力、丙二醛(MDA)、过氧化氢(H2O2)和还原型谷胱甘肽(GSH)的含量.结果 与正常组比较,帕金森病模型组大鼠的逃避潜伏期明显延长(P<0.01),跨越平台次数明显减少(P<0.01);经半夏总生物碱治疗组,大鼠逃避潜伏期明显缩短(P<0.05,P<0.01),跨越平台次数明显增加(P<0.01).帕金森病模型组大鼠脑皮质及血清中MDA、H2O2的含量增加,SOD活性及GSH的含量降低(P<0.01);经半夏总生物碱治疗组,脑皮质MDA、H2O2的含量显著减少(P<0.01),皮质GSH、SOD含量显著增加(P<0.01);半夏总生物碱给药组中低浓度组、中浓度组血清MDA的含量无统计学意义(P>0.05),高浓度组血清MDA含量下降(P<0.01),各治疗组中血清H2O2含量明显下降(P<0.01),血清GSH含量显著增加(P<0.01).结论 半夏总生物碱具有改善学习记忆能力,对抗大鼠神经系统的退行性变有一定的作用,可能通过改变帕金森病模型大鼠皮质部分及血清SOD、GSH的含量,而抑制了MDA和H2O2的产生.  相似文献

三叶半夏组培快繁与优良单株的选育   总被引:5,自引:1,他引:4  
本文利用组织培养技术实现了三种不同叶型三叶半夏的快速繁殖,并通过光合生理特征的比较对其优良单株进行了选育。结果表明:半夏能在分化培养基MS+6-BA 1.0 mg/L + NAA 0.2 mg/L+蔗糖3%+琼脂5.0 g/L和离体块茎诱导培养基(1/2 MS+IBA 0.03 mg/L+ NAA 0.01 mg/L+蔗糖5%+琼脂5.0 g/L+AC 0.3 g/L)上快速扩繁,继代繁殖系数为1:6.23,培养3个月后块茎的平均直径达0.88cm。 对其光合生理特征的研究结果表明:三叶半夏是一种典型的阴生植物,其光饱和点为700μmol·photons/s·m2;三种叶型的三叶半夏都存在着明显的光抑制现象,其中柳叶型最明显。本研究所选育的11个单株后代中,以桃叶2号(T2)的净光合速率和产量最高,柳叶2号(L2)对强光的耐受力最强。经过6个月的扩繁,本研究获得优良单株T2和L2离体块茎各1万颗。在温州乐清实验基地中经过1年的大田种植后得优良单株半夏块茎各100kg。  相似文献

在同一栽培条件下,对主要引自长江中下游地区的15个半夏〔Pineliaternata(Thunb.)Breit.〕居群的16个主要形态性状进行模糊聚类分析,在置信水平λ=0.6764时,将15个居群划分成4个类型:(1)双珠芽类型:主要形态特征为叶柄上均具双珠芽,但叶型和块茎形状变异较小;(2)普通类型:主要形态特征为叶柄上均只着生单珠芽,但叶型和块茎形状变异较大;(3)长茎类型:主要形态特征为叶柄上具单珠芽,但着生位置较低,块茎呈矩圆形;(4)复合类型:主要形态特征为叶柄上具单珠芽,但居群内常有双珠芽个体出现。  相似文献

探讨高温胁迫下半夏倒苗前后的组织学结构的变化,为揭示半夏倒苗的生物学机理提供参考依据。在半夏植株高15cm左右时,给予(32±1)℃的高温胁迫,于不同胁迫天数取半夏叶柄,经爱氏苏木精染液染色,用石蜡切片法制片,在显微镜下观察并照相。结果表明,随着高温胁迫时间的延长,半夏叶柄的维管组织结构被破坏越来越严重,细胞壁、细胞膜破裂,原生质外渗,核膜破裂,核仁溢出,有的细胞内核仁消失。  相似文献
